Superior Extrahepatic Tropism

Unlike traditional LNPs that get trapped in the liver, RJH reagents bypass the hepatic barrier and reach essential targets such as bone marrow.

Leu‑Fect-formulated LPNPs drive potent extrahepatic siRNA delivery, with elevated siRNA levels in bone marrow, spleen, and lung 24 hours after IV dosing in BALB/c mice (n=5).


Sustained Therapeutic Retention

Why inject constantly when your delivery vehicle can retain the payload? While LNP-delivered siRNAs are cleared rapidly from the body, lipopolymers protect and retain therapeutic nucleic acids in target tissues for over a week after a single injection.

NCG mice (n=5) treated with Leu-Fect/siRNA maintained robust, therapeutic siRNA levels in liver, bone marrow, spleen, and lung after 7 days. siRNA levels in LNP-treated mice were almost undetectable in the same timeframe. 


Better and Sustained Efficacy

Optimizing knockdown effectiveness together with maintaining prolonged cytotoxicity is crucial for cancer therapeutics. ALL-Fect, the RJH reagent, is perfectly suited for this task.

In A549 cells (pulmonary epithelial cells) treated with 60 nM siRNA, RJH reagent delivery of STAT5- or SURVIVIN-targeting siRNAs reduced cell viability to 35-50% on day 3 and maintained this low viability through 9 days. Standard LNP formulations (LNP1 and LNP2) showed poor efficacy.
